Aldehyde dehydrogenase 1 family member A1 (ALDH1A1), also known as retinal dehydrogenase 1, belongs to the aldehyde dehydrogenase enzyme family, which is involved in the metabolism of alcohol. ALDH1A1 is predominantly expressed in the epithelium of testis, brain, eye, liver, kidney, as well as neural and hematopoietic stem cells. Reportedly, high ALDH1A1 expression is found in solitary fibrous tumor (SFT) and hemangiopericytoma (HPC), compared to meningiomas and synovial sarcomas. Recognizes a protein of 80 kDa-90 kDa, identified as CD36. It is expressed on platelets, monocytes and macrophages, microvascular endothelial cells, erythrocyte precursors, mammary epithelial cells, and some macrophage derived dendritic cells. CD36 acts as a receptor for thrombospondin (TSP), collagen types I, IV and V, P. falciparum malaria-infected erythrocytes, and sickle erythrocytes. It also functions as a scavenger receptor, mediating macrophage uptake of oxidized low-density lipoprotein (LDL) and recognition of apoptotic polymorphonuclear leukocytes (PMN). CD36 plays a role in platelet aggregation, macrophage foam cell development, inflammation, and the tissue ischemia observed in sickle cell disease and cerebral malaria. This MAb recognizes a protein of 36 kDa, identified as ICOS-L. It is a ligand for the ICOS receptor that initiates T and B cell proliferation and cytokine secretion. ICOS-L interactions play an essential role in T cell-dependent B cell activation in peripheral lymphoid organs such as spleen and lymph nodes. ICOS-L protein is present in myeloid leukocytes, and by Northern blot there are 2. 4, 3. 0, and 7. 0 kb transcripts in brain, heart, kidney, and liver, with lower expression in colon and thymus, and a 1. 1 kb transcript in leukocytes. Tumor necrosis factor alpha (TNF), granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-4 (IL-4) enhance its expression. This antibody recognizes a 32 kDa protein, which is identified as homodimer of galectin-13 (also known as PP13). Galectins are a family of soluble beta-galactoside-binding lectins that modulate cell-to-cell adhesion and cell-to-extracellular matrix (ECM) interactions and play a role in tumor progression, pre-mRNA splicing and apoptosis. One such member, galectin-13, is a 139 amino acid protein that contains one galectin domain. Possessing lysophospholipase activity, galectin-13 exists as a disulfide-linked homodimer. Galectin-13 is suggested to have a developmental role in the placenta and may display immunobiological activity in feto-maternal blood-spaces. Recognizes a 53 kDa protein, identified as the Ornithine Decarboxylase (ODC-1). ODC is the initial and rate-limiting enzyme in the biosynthetic pathway of polyamines and is involved in the conversion of ornithine to putrescine. The biological activity of ODC-1 is rapidly induced in response to virtually all agents known to promote cell proliferation including hormones, drugs, growth factors, mitogens, and tumor promoters. Reportedly, ODC mRNA levels are elevated in lung carcinomas as well as in colon adenomas and carcinomas. ODC activity in colorectal carcinomas is greater than those in adenomas and normal mucosa. This Ab reacts with the rod domain of human cytokeratin 19 (CK19), a polypeptide of 40 kDa. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. NGFR is expressed in various neural crest cells and their tumors such as melanocytes, melanomas, neuroblastomas, pheochromocytomas and neurofibromas. Reportedly, anti-NGFR is a reliable marker for desmoplastic and neurotropic melanomas.
